Abstract: The assembly of spacecraft in clean room environment is essential for the prevention of forward contamination. In this study, both high-throughput sequencing and culture-based method are used to study the microbial community structure and diversity in the AIT (Assembly, Integration and Test) center of the spaceflight. The result of high-throughput sequencing analysis show that in the AIT center the dominant genus of airborne bacteria is Bacillus (78.47%±1.59%), and the dominant fungi is Tremellales (8.97%±0.93%). However, culture-based analysis indicates that the Staphylococcus is the dominant bacteria, and no fungal colony is observed in the AIT center. The microbial diversity is evaluated by chao1 index, Simpson index and Shannon index, and the results show that the bacterial diversity level is higher than that of fungal in the AIT center.
